Experiment No. 1
Aim of Experiment: - VDRL Test By Card Method
Principle: - Non-treponemal antigen (Cardiolipin-Cholesterol-Lecithin) is used to
detect the presence of “reagin antibodies” ( IgM and IgG antibodies to lipoidal
material released from damaged host cells as well as to lipoprotein-like material,
and possibly cardiolipin released from the treponemes) in patient’s serum.

Requirement: - VDRL Test Kit contain


 Non treponemal slide
 Antigen solution
 Capillary tubes
 Rotator
Procedure: -
1. Take all the requirement of the test.
2. Then take a sterilize card of VDRL TEST.
3. Then put 50ml of serum sample in one well.
4. Then add VDRL antigen in the sample.
5. Mix it well then keep it on rotator at 180 rpm .
6. Then observe agglutination.

Result: -
Negative: - agglutination not seen
Positive: - agglutination seen
Experiment No. 2
Aim of Experiment: - WIDAL Test By Card Method
Principle: - The main principle of widal test is that if homologous antibody is present
in patients serum, it will react with respective antigen in the reagent and gives visible
clumping on the test card and agglutination in the tube. The antigens used in the test
are “H” and “O” antigens of Salmonella Typhi and “H” antigen of S. Paratyphi. The
paratyphoid “O” antigen are not employed as they cross react with typhoid “O” antigen
due to the sharing of factor 12. “O” antigen is a somatic antigen and “H” antigen is
flagellar antigen.

Requirement: - Widal test kit contain


 Antigen (O, H, OH)
 Widal test card
 Rotator
 Capillary tube
Procedure: -
1) First take all the requirement.
2) Than put 20 micro liter serum sample into 3 wells.
3) Than add O antigen into first well, H antigen into second well
and OH antigen into third well.
4) Mix them well with the help of capillary tube.
5) Put the plate on the rotator at 120 RPM for 2 minutes.
6) Observe the agglutination.

Result: -
Positive: - if the agglutination seen in the wells it means the
test is positive.
Negative: - if the agglutination not seen in the wells that
means the test is negative.
Experiment No.: - 3
Aim 0f Experiment: - MALERIA TEST BY CARD METHOD
Principle: - Malaria parasites produce proteins called antigens. RDTs
detect these malarial antigens in a person’s blood. If malaria antigens are
present, the test will be positive. If malaria antigens are not present in the
person’s blood (RBCs) the test will be negative. Different types of RDTs
detect different antigens of malarial parasites.
Requirement: - Malaria kit contain
 Test card strip
 Sample running buffer solution
 Pipette

Procedure: -
1) Take all the requirements.
2) Take the card strip.
3) Add 5microliter of patient whole blood with the help of micropipette in
the sample well.
4) Then add 4 drops of buffer solution in the card well.
5) Wait 15 minutes for result.
Result: -
I. If the line of plasmodium vivex and plasmodium falciparum
line’s color will change that means the test is positive.
II. If the only control line’s color will change that means test is
negative.
Experiment No. 4
Aim of the experiment: - C Reactive Protein Test (BY
Card Method)
Principle: - This is a slide agglutination test for the qualitative and semi quantitative
detection of C-Reactive Protein (CRP) in human serum. Latex particles coated with
goat IgG anti-human CRP are agglutinated when mixed with samples containing CRP.
When latex particles coated with human anti-CRP are mixed with a patient’s serum
containing C – reactive proteins, this results in visible agglutination with in 2 minutes.

Requirement: - CRP test kit contain


 Test Slide
 Anti CRP reagent
 Rotator
Procedure: -
1. First take all the equipment.
2. Then take the test card.
3. Put 50 micro liter of patient serum with the help of micro pipette in the
wells.
4. Then add the CRP reagent 2 to 4 drops into the patient serum.
5. Keep the plate on the rotator for 2 minutes at the 180 RPM.
6. Observe for the agglutination.

Result: -
 If the agglutination will not seen that means the test is negative
 If the agglutination will seen that means the test is positive.
Experiment No. 5
Aim of the experiment: - HBSAG BY CARD METHOD
Principle: - The One Step HBsAg Rapid Test is based on the principle of
chromatographic immunoassay for qualitative detection of the surface antigen of
hepatitis B virus (HBsAg) in human whole blood, serum, and plasma samples.
Monoclonal and polyclonal antibodies are employed to identify HBsAg specifically.

Materials Provided with HBsAg Rapid Test Kit:

1. HBsAg Test device


2. Sample diluents
3. Instructions for use

Procedure: -
I. First take all the requirement.
II. Then add 10 micro liters of patient serum into the well.
III. Then wait for 10 minutes.

Result: -
There are two lines in the card test. First is the control line and second one is
test line.
 If only control line will appear that means the test is negative.
 If the both control and test line will appear that means the test is positive.
Experiment No. 6
Aim of Experiment: - HIV BY CARD METHOD
Principle: - this test works by evaluating the blood of the patient. A specimen of
serum must be provided in order for the test to be accurately completed. This
specimen is then tested for the presence of HIV antibodies. These antibodies bond
with the gold conjugated antigens that are provided in the test kit. If HIV antibodies
are present in the specimen are red line appears in the test zone to indicate that the
individual has tested positive for HIV. The control zone must appear red every time
the test is performed.

Requirement: -
 Test card
 Buffer solution
 Pipette
 Conjugate

Procedure: -
1) First take all the requirement.
2) Then add 2 drop of buffer solution into the well.
3) Add 10 micro liters patient serum into the well.
4) Then add conjugate solution which is provided in the kit.
5) Then again add 1 drop of buffer solution into the well.
6) Wait for the 10 minutes.
Result: -
 If only the control line will appear that means the test is negative.
 If the control and test both line appear that means the test is positive.
