1 Introduction

Freeze-Drying is an extremely useful technique for the biologist as it enables him to preserve valuable but
unstable preparations, and many types of living organisms, in perfect condition for long periods at moderate to
high temperatures. In most biological laboratories freeze-drying is now a routine procedure and has been of
great benefit to pharmaceutical industry as a stage in manufacture, particularly in the concentration of heat-
sensitive solution, and in the preparation of stable, highly soluble ethical products.
The Cleanvac 8(12) freeze dryer is complete system for pre-freezing, primary drying and secondary drying. Three
petri dishes of φ230mm can be inserted in the cylindrical style transparent acrylic chamber, which is installed on
the 8(12) liters capacity cold trap. Heater plates(optional) can be attached on the lack for petri dishes to reduce
the drying procedures. The pirani gage is installed in the system to measure the vacuum pressure and pt-100
temperature sensors are used to measure the temperature of cold trap, specimen(optional) and heater
plates(optional). The whole freeze drying procedures are digitally monitored and controlled with PIC standalone
microcomputer and all the measured values are displayed immediately on the LCD monitor. The drying time can
be adjusted by internal timer and after the setting time is passed, the buzzer is beep and all the drying procedure
is terminated. In order to measure the remaining moisture in the specimen, the vacuum pump is turned off for
two minutes in every 3 hours and checks the vacuum pressure variation. If the vacuum pressure is maintained in
the specified level, the drying process checking algorithm gives the message of “drying process is terminated”on
the LCD monitor. But the vacuum pump is running continuously to perform the secondary drying procedure.
Instead of clear chamber, Flask manifold can be attached on the cold trap(optional) that it is possible to dry the
specimen with using the various sizes of flasks.

What Freeze-Drying is?


Freeze-drying is intended to preserve wet substances, or solution in water, that are usually, but not always, of
biological origin. The product is frozen and then exposed to an atmosphere of low relatives humidity when the
contained ice sublimes-that is, it changes directly from the solid to the vapour state without melting, thereby
avoiding the chemical, physical and enzymic changes usually associated with other forms of drying. In practice
the necessary low relative humidity is achieved by conducting the process under vacuum.
Freeze-drying is conventionally divided into three stages: Pre-freezing, sublimation drying under vacuum, known
as primary drying, followed by desorption or secondary drying. During primary drying the ice does not evaporate
simultaneously from all parts of the material, but a frozen interface continuously moves away from a dry outer
boundary. In secondary drying the few percent by product weight of residual, unfreezable or partially combined
moisture, which remains after primary drying, is reduced to an optimum value (commonly 1 %) for product stage.

Benefit
Cleanvac 8(12) improves lab productivity. They are the only reliable way to lyophilize all types of samples. The
user friendly designed controller gives step by step procedures for the system operation. The whole procedures
are digitally monitored and controlled with standalone PIC microcomputer. All the measured values such as
temperatures of trap, specimen and heater plates, and vacuum pressure are displayed on LCD monitor. The
freezedrying time can be adjusted by timer function.
Freeze-drying or lyophilization requires pre-frozen samples. The oversize cold trap of Cleanvac 8(12) can freeze
the specimen without using extra pre-freezing apparatus.

Most freeze-dryers cannot process solutions that are strongly acid, basic, or saline, thereby allowing vapors to
reach and corrode the vacuum pump. The ultra-low-temperature refrigerated trap and the chemical trap catch
vapors and prevent them from reaching the vacuum pump. An oil filter recirculator purifies the oil in the vacuum
pump.

3 Installation

3-11. Safety
Please follow safety measure as describing the manual in order to protect any possible danger situations as
damaged product, failure, and casualty.

1) Install the machine on the flat place with horizontality.


2) Check rated voltage before connected with machine and power.
3) Use Hanil’s spare parts and accessories.
We don’t have responsibility for damaged products and accident if the user is using not recommending spare
parts and accessories.
4) In case of pathogenicity, toxicity, radiological material, please check necessary precaution and inspect enough a
property of matter.
5) If the machine is contaminated by pathogenicity, toxicity, radiological material, eliminate contaminant
completely. And please do necessary action as ventilation or isolation of centrifuge.
6) Do not use possible substance for volatile gas and explosiveness gas.
7) Do not remove flask forcibly during running the machine.

3-2. General checking details


Package Examine whether the product has damage for package if it is arrived. If there
are damage or lack of spare parts, contact with Hanil Science Industrial
company immediately. Contact points were written on the package box and
user manual cover.

Stable and Flat The product should be set up on the hard and flat floor. In case the product
place is set up in inclined place, it will cause defective machine.

Space The necessary space requirements can be found under dimension in the
requirements technical data chapter. The product must be set up in a suitable space, so
and proper that it is stable. For proper circulation of air, the product should be
circulation of established on the place that it defined with 30cm of both side and 30cm of
air space on back. If draft hole on the back of case is blocked by hiding with
clothes or is blocked from other device, air circulation is difficult and
especially avoids place that dust happens much.
Temperature, The product is influenced in temperature or external environment of humidity
Humidity because is operated by highly electronic control system. Do not establish
near direct ray of light or heating utensil. Proper temperature, humidity
should be kept.

to occur acid gas or chlorine gas, some materials will be corrosion possibly.

3-3. General checking details


CHECK 1: remaining oil in vacuum pump
Check remaining oil whether it is enough or not. Oil level will be located between Max. and Min. and
recommending oil level will be max. 5mm. if oil level is located in within 25mm height, the oil will be replenished.
And when the pump is running, oil level will be going down as normal situation. And please check oil clarity with
alien substance.
CHECK 2: chemical trap
You can check cartridge replacement with weight and visual inspection whether or not. If inside of cartridge filling
material color is changed, cartridge should be replaced. And if the weight of cartridge is increasing twice or treble
than previous one, cartridge should be replaced. Also, in case accumulating of solvent as hazardous material
should be replaced all the time.

3-4. Electric requirements details


Please use rated voltage registered in label of product for normal operation of device. Please inquire in the
local supply of electric power company or customer support center in case of confirm whether engaged rural
districts fit in rated voltage of product.

Plug that protection ground connection pin is to product is offered, and this plug can insert to ground
connection way electricity outlet for safety. Exchange this plug for ground connection in case of do fit
to busy outlet. This time, please take care lest should get shocked, and inquire to professional
electrical engineer according to need. Ground connection way adaptor plug does not use absolutely to
electricity outlet that ground connection monad is not.

When errors happen, to cancel power connection, the product must establish emergency switch to
drop in the machine. Laboratory way out lateral or outside is good.

In order to protect damaged compressor by unstable condition of delivery, please wait 4 hours before
turning on switch after installation.

3-4-1. Electric safety information


Use Hanil’s power cable to offer with other equipments.
• Put the main power code with neck of electric receptacle which is becoming ground connection.
Avoid multi receptacle.
• If product’s ground wire is connecting wrong, it will cause electric shock.
• Install the product to avoid stepping the power code.
• Do not put anything on the power code.
• Avoid being block in ventilating opening.
• Do not put anything into slot or hole in the product.
• If following situation is occurring, turn off main power and unplug power code.
- Strange sound or bad smell in the product.
- Damage or abrasion for power code.
- In case cutting the circuit breaker in the surface of a wall, fuse or other safety device.
- Spill the liquid in the machine.
- Damage for some part of machine.

Start <Circuit Breaker> Switch the circuit break on at the back of the instrument

Press any button down to go for set-up mode

<Cold Trap On> Cool Cold Trap down with the set temperature (-45℃ or -85℃) in advance
and make a next progress

Continuous mode Condition

As mode for setting the timer, press “UP” or “DOWN” button down to
change the temperature value. 24Hr is the maximum setting time, the timer
changes to the continuous mode more than 24Hr.
Operating time can be controlled by starting the vacuum pump. Once
operating time finishes, every system function including alarm stop. At this
moment, alarm function is cancelled by pressing any key down

5-2-7. Operation sequence in case of using Manifold block & flask

• Switch the main power on


• Put all the flasks with the sample into the cold trap
• Switch the cold trap on and freeze the sample for more than 4 hours
• It takes about an hour or so for cold trap to be frozen by -80℃
• Put the flasks in the manifold
• Switch the vacuum pump on
• Wait till the vacuum pressure drops down (Normally 0.1 Torr)
• Return the manifold handle of each flask to the open. It normally takes 10~20hours for freeze-drying
• Moisture or ice should be removed on the surface of the flasks to reduce the time for freeze-drying. Moisture
or ice does not come out once freeze-drying reaches to the final stage
• Automatic process-watch Algorism stops the vacuum pump for 2 minutes every 3 hours and checks the
changing amounts of the pressure. When the change of the pressure keeps within the regular level, the
process-watch Algorism sends the message of the end of freeze-drying to the LCD monitor
• After freeze-drying finishes, shut the cork of the manifold and separate the flasks from the chamber of the
manifold
• Open the ventilating valve

5-2-8. Operation sequence in case of using the clear chamber & Petri dish

• Switch the main power on


• Put the whole rack of the Petri dish with the sample into the cold trap
• Switch the cold trap on and freeze the sample for more than 4 hours
• It takes about an hour or so for cold trap to be frozen by -80℃
• Put the drying chamber on the cold trap and put the Petri dish rack into the inside
• Close the cover of the drying chamber and switch the vacuum pump on
• It normally takes 10~20hours for freeze-drying
• Automatic process-watch Algorism stops the vacuum pump for 2 minutes every 3 hours and checks the
changing amounts of the pressure. When the change of the pressure keeps within the regular level, the process-
watch Algorism sends the message of the end of drying to the LCD monitor
• Open the ventilating valve

6 Maintenance

6-1. Cleaning

6-1-1. Instrument
• Wipe the external surface with a soft cloth soaked with soapy water. Then, wipe it with a dry cloth to dry the
instrument
• Do not use chemicals including alcohol, benzene and thinner which can damage the instrument. Wipe the
instrument with a soft cloth soaked with a good neutral detergent
• Do not scratch the external surface during cleaning.
Do not use metal sponge.
Do not scratch the instrument during the movement of the instrument
The scratch on the surface will rust the instrument
• If the user leaves it wet for a long time and it becomes rusty, remove the rust with a neutral detergent and wipe
it with a dry cloth

6-1-2 Refrigeration
• Protect a radiator from the contaminant including dust and dirt to keep the efficiency of the refrigeration
system. Otherwise, it will reduce the function of the refrigeration. Clean the refrigeration with a brush or
vacuum it at least once three months

6-2. Maintenance

6-2-1 General checking details


• Control board: check the function and condition of connection between the support bolts
• Connector: Check whether it is loosened or it is inserted properly
• Manifold valve: Check whether the inserting area of the Flask is damaged or not

6-2-2 Storage
• Remove the contaminant on the surface of the instrument after using
• Surely switch the power off and clean the door (lid)
• Wipe the main body and rotor with a warm neutral detergent
• Clean the flask periodically
• Keep out of direct sunlight
